Overview

Teashirt zinc finger homeobox 3 (TSHZ3) is a zinc-finger homeobox transcription factor essential for development of urinary tract smooth muscle, respiratory rhythm neural circuits, and regulation of gene expression through chromatin association and histone deacetylase recruitment. It interacts with other transcription factors such as SOX9 and myocardin to regulate differentiation, and loss or mutation of TSHZ3 leads to congenital malformations of the kidney and ureter, as well as neural defects resulting in breathing abnormalities. Heterozygous deletion or reduced expression of TSHZ3 in humans is linked to autism spectrum disorder, intellectual disability, and renal tract abnormalities. In certain cancers, epigenetic silencing of TSHZ3 may promote tumor growth. While TSHZ3 is under investigation for its roles in disease, no drugs currently target this protein directly, and its pleiotropic developmental functions create safety challenges for direct pharmacological intervention.
