The Tec family tyrosine kinases represent the second-largest family of non-receptor tyrosine kinases, characterized by a unique Pleckstrin Homology (PH) domain at the N-terminus. This family consists of five members: TEC, BTK (Bruton's tyrosine kinase), ITK (Interleukin-2-inducible T-cell kinase), BMX (Bone marrow kinase on the X chromosome), and TXK (also known as RLK). These kinases are predominantly expressed in hematopoietic cells and serve as critical mediators of signaling downstream of antigen receptors, such as the B-cell receptor (BCR) and T-cell receptor (TCR), as well as cytokine and integrin receptors (PMID: 25135138; UniProt P42680). Clinically, BTK is the most significant member of the family, playing a vital role in B-cell maturation and survival. Dysregulation of Tec family kinases is linked to various pathologies, including B-cell malignancies like chronic lymphocytic leukemia (CLL) and mantle cell lymphoma, as well as autoimmune disorders like rheumatoid arthritis and systemic lupus erythematosus (StatPearls NBK542211). Therapeutic strategies primarily involve small-molecule inhibitors that target the ATP-binding site of the kinase domain. While highly effective in oncology, challenges include the development of resistance mutations (e.g., C481S in BTK) and off-target toxicities such as atrial fibrillation and bleeding, which are often linked to the inhibition of other Tec family members or related kinases like EGFR (PMID: 30736293).
