Overview

Tectonic family member 1 (TCTN1) is a transmembrane and secreted protein that is part of the tectonic family, involved in the formation and function of primary cilia—cellular organelles essential for developmental signaling pathways, including the Hedgehog pathway[1][2]. TCTN1 has been implicated in regulating ciliary membrane composition and serves critical roles in vertebrate embryogenesis, particularly in neural tube patterning and organogenesis[1]. Dysregulation and overexpression of TCTN1 are associated with several cancers (including glioblastoma and oral squamous cell carcinoma), where it promotes tumor cell proliferation, migration, and invasion[1][2]. It is also involved in human genetic disorders known as ciliopathies (e.g., Joubert and Varadi syndromes), and may act as a novel oncogenic driver and a potential therapeutic target in select tumors[1][2]. Currently, no clinically approved drugs specifically target TCTN1, but its expression may have clinical value as a biomarker for tumor aggressiveness and disease progression[1][2].
