TEK receptor tyrosine kinase (TIE2) is a cell surface receptor expressed primarily in endothelial cells and hematopoietic stem cells. It plays a critical role in vascular development, angiogenesis, and communication between endothelial cells and smooth muscle cells. Activation by angiopoietins, particularly Ang1, initiates signaling pathways that regulate endothelial cell survival, proliferation, and vessel integrity. Mutations leading to constitutive activation of TIE2 are associated with venous malformations, and TIE2 is also implicated in tumor angiogenesis.
