TEK tyrosine kinase (Tie2)

Target
Tie2
Molecular classification
Receptor Tyrosine Kinase, Receptor
01

Overview

TEK tyrosine kinase (Tie2) is a receptor tyrosine kinase primarily expressed on endothelial cells. It is essential for the development, remodeling, and maintenance of the vascular system. Tie2 mediates communication between endothelial cells and smooth muscle cells, which is necessary for blood vessel formation (angiogenesis) and maintaining vessel integrity. Mutations or dysregulation in TEK/Tie2 are associated with multiple cutaneous and mucosal venous malformations (VMCM) and tumor angiogenesis.

TIE2CD202BAngiopoietin-1 receptor
02

Mechanism of action

Tie2 inhibitors and antibodies target the receptor to block its activation and downstream signaling, primarily aimed at reducing tumor angiogenesis. Agonists may also be used to promote vascular stability.
