Overview

Tie2 (TEK Tyrosine Kinase Receptor) is a receptor tyrosine kinase primarily expressed on endothelial cells, playing a critical role in angiogenesis, vascular stability, and remodeling. It is activated by Angiopoietin-1 (Ang1), leading to downstream signaling through pathways such as PI3K-Akt and MAPK/ERK, promoting endothelial cell survival, proliferation, and migration. Angiopoietin-2 (Ang2) acts as a context-dependent antagonist or partial agonist. Tie2 is a key therapeutic target in cancer, with antiangiogenic therapies aimed at inhibiting its activity to suppress tumor growth. Mutations in TEK are associated with venous malformations.
