Telomere repeat-binding bouquet formation protein 1 (TERB1) is a meiosis-specific protein that acts as a molecular scaffold, critical for tethering telomeres to the inner nuclear membrane during prophase I of meiosis. It forms a stable complex with TERB2 and MAJIN, mediating the association of telomeric DNA (shelterin complex and TRF1) with the nuclear envelope. This NE attachment is required for homologous chromosome pairing, synapsis, and the progression of meiotic division. Disruption in TERB1 or its interacting partners abolishes telomere-NE attachment, leading to synapsis defects and infertility in mammals. TERB1 is not a classical therapeutic target such as a receptor, enzyme, or transporter, but rather a structural/organizing protein essential for reproductive cell meiosis[1][2][3].
