Telomere repeat-binding bouquet formation protein 2 (TERB2) is a meiosis-specific, telomere-associated protein central to chromosome dynamics in germ cells. TERB2 is a component of the tripartite meiotic telomere complex (MTC), along with TERB1 and MAJIN, which is vital for the attachment of telomeres to the inner nuclear membrane during meiotic prophase I. This tethering is essential for homologous chromosome pairing, synapsis, and normal progression through meiosis. Disruption of TERB2 function, or loss-of-function mutations, abrogate telomere-nuclear envelope attachment, leading to meiotic failure, germ cell maturation arrest, and clinically, male infertility (notably non-obstructive azoospermia). TERB2 does not currently represent a therapeutic target in the contexts of pharmacology, oncology, or chemical biology, nor are any drugs known to modulate its function in clinical or preclinical settings.
