Overview

Temozolomide resistance associated STAT3 regulator 1 (TMZR1) is a non-coding enhancer RNA transcribed from the STAT3 locus in human cells. It acts as a key regulator of STAT3 gene expression, particularly in glioblastoma, where elevated TMZR1-eRNA levels are associated with increased resistance to the chemotherapy agent temozolomide. Research indicates that knockdown of TMZR1-eRNA decreases STAT3 transcription and protein levels, sensitizing cancer cells to temozolomide and significantly reducing cell viability. TMZR1-eRNA is expressed at much lower levels in normal brain and peripheral blood cells, suggesting its role as a glioblastoma-specific regulator. Modulation of TMZR1-eRNA thus represents a promising, tumor-selective approach to overcoming temozolomide resistance with potentially fewer systemic side effects than direct inhibition of STAT3 protein.
