Overview

Tenascin-C (TNC) mRNA is the messenger RNA transcript that encodes Tenascin-C, a large hexameric extracellular matrix (ECM) glycoprotein (UniProt: P24821). While Tenascin-C is essential during embryonic development, its expression in healthy adult tissues is highly restricted, appearing primarily during wound healing or in response to mechanical stress. However, TNC mRNA is significantly overexpressed in various pathological conditions, including solid tumors, chronic inflammation, and fibrotic diseases (PubMed: 30333295). In the context of oncology, the resulting protein promotes tumor cell proliferation, epithelial-mesenchymal transition (EMT), and angiogenesis, making the mRNA an attractive target for silencing therapies. Therapeutic strategies such as siRNA (e.g., STNM01) aim to degrade TNC mRNA to reduce the protein's contribution to disease progression and tissue remodeling (PubMed: 21854181).
