Overview

Tenomodulin is a tendon- and ligament-specific type II transmembrane glycoprotein found predominantly in tenocytes, ligamentocytes, tendon stem/progenitor cells, and periodontal ligament cells. It plays central roles in tissue maturation, cell proliferation, and maintenance of tendon and ligament homeostasis. Tenomodulin contains a conserved BRICHOS domain and an anti-angiogenic, cysteine-rich C-terminal region analogous to chondromodulin-1. Its expression is largely restricted to hypovascular tissues, and its loss impairs tenocyte proliferation and collagen matrix organization. Genome-wide associations implicate TNMD in metabolic and neurodegenerative diseases, but its principal characterization is within connective tissues. Although no drugs currently target this protein, it is intensively studied as a potential regenerative marker and regulator in tendon biology

Safety considerations

No major therapeutic safety concerns described due to the absence of direct drugs targeting tenomodulin. However, knockout models show impaired tendon maturation, reduced cell density, and abnormal collagen accumulation, indicating its essential role in tendon integrity and function
