Overview

Tensin-3 (TNS3) is a member of the tensin family of focal adhesion proteins that physically link the actin cytoskeleton to integrin receptors at cell-matrix adhesion sites[1][3][4]. It contains N-terminal actin-binding, C-terminal Src homology 2 (SH2), and phosphotyrosine-binding (PTB) domains, allowing it to bind both actin filaments and the cytoplasmic tails of certain beta integrins[3][4]. TNS3 acts as a scaffold protein involved in cell migration, adhesion, and signal transduction, especially in response to growth factors such as EGF[1][2][4]. It regulates Rho GTPase pathways through interactions with the RhoGAP DLC1 and the GEF DOCK5, and participates in the assembly and organization of podosome belts in osteoclasts[1][2][4]. Tensin-3 supports cell proliferation, especially in mesenchymal stem cells and certain cancer cell lines[1][2]. Aberrant expression of tensin-3 is linked to cancer progression and metastasis, and it is a recently identified substrate of MALT1 protease, affecting B cell adhesion and immune functions[3].
