Overview

TEPSIN adaptor related protein complex 4 accessory protein (TEPSIN) is an accessory subunit of the AP-4 protein complex, which forms a non-clathrin coat on vesicles departing the trans-Golgi network. TEPSIN interacts with specific domains of AP-4 subunits and is thought to participate in the proper assembly, cargo sorting, and distribution of vesicular proteins, particularly in neuronal cells. While the AP-4 complex is essential for neural development and mutations in its subunits cause severe neurological disorders (such as hereditary spastic paraplegia), TEPSIN’s exact biological role remains poorly defined. There is no evidence that TEPSIN itself is directly implicated in human disease or targeted by any therapeutic agents, but its association with AP-4 suggests an important auxiliary role in protein trafficking and possibly neuronal functions
