Terminal Nucleotidyltransferase 4B (TENT4B), also known as TUT3 or PAPD5, is an enzyme that catalyzes the addition of nucleotides, preferentially ATP and GTP, to the 3' poly(A) tail of RNA molecules. This activity influences mRNA stability, quality control, and other RNA metabolic processes. TENT4B is involved in post-transcriptional regulation by modifying mRNA tails, shielding them from rapid deadenylation and degradation. It is found in the cytosol, nucleolus, and plasma membrane and is a component of the TRAMP complex. Variants in TENT4B are associated with diseases involving defects in mRNA processing or stability, and chemical inhibition shows promise in rescuing telomerase function and hematopoiesis under certain pathological conditions.
