TESK2 is a dual specificity protein kinase (phosphorylating serine/threonine and tyrosine residues) with structural similarity to the LIMK/TESK family and is most closely related to TESK1. It phosphorylates substrates such as cofilin, influencing actin cytoskeleton remodeling and is predominantly expressed in testis and prostate. In rodent models, TESK2 is crucial during meiotic and early spermiogenic stages, suggesting a role in male germ cell development and function. Disease associations include developmental and epileptic encephalopathy 28, with additional roles possible in cell signaling, cytoskeletal organization, and potentially cancer, although targeted therapies and clinical biomarkers are unreported.
