Overview

Testis-expressed protein 12 (TEX12) is a coiled-coil structural protein essential for meiosis, where it acts as a core component of the synaptonemal complex, facilitating homologous chromosome synapsis through fibrous assembly with SYCE2[1][2][4]. TEX12 can also localize to centrosomes, both in meiosis and, aberrantly, in some cancers—where its ectopic expression leads to centrosome abnormalities and is associated with poor clinical outcomes[1][3]. TEX12 exists primarily as a dimer through its central α-helical domain and mediates higher-order assembly of the synaptonemal complex. Aside from its meiotic role, its oncogenic involvement derives from deregulated expression in tumors, making it a cancer-testis antigen and potential biomarker, but no direct drug interactions or specific mechanisms of pharmacological action are established at this time[1][3][4].
