Testis-expressed transcript, Y-linked 14 (TTTY14), is a long non-coding RNA (lncRNA) located on the Y chromosome (Yq11.222) that does not code for a protein[1][3]. It shows testis-specific expression but is upregulated in various cancers, notably in testicular germ cell tumors (TGCT), and its high expression is associated with poorer survival in TGCT patients[1][2]. Experimental data indicate that TTTY14 promotes proliferation of TGCT cells, with knockdown reducing cell viability and growth[1][2]. The expression of TTTY14 is regulated through copy number variation and DNA methylation[1]. TTTY14 appears to influence the tumor immune microenvironment, being correlated with higher immune dysfunction and altered abundance of immune cell types (decreased B cells, CD8+ T cells, and macrophages, increased CD4+ T cells)[1]. Elevated TTTY14 may reduce sensitivity to multiple anti-cancer therapies and is also being studied as a prognostic and predictive biomarker for cancer therapy and immunotherapy response[1]. TTTY14 is not a receptor, enzyme, or therapeutic target in the typical sense, but rather a noncoding RNA with roles in gene regulation and cancer biology[1][3]. There are currently no known drugs that directly target TTTY14, nor established mechanisms of action for pharmacological modulation[1][2][3].
