Testis-specific Y-encoded-like protein 2 (TSPYL2) is a nucleosome assembly protein found mainly in the nucleolus, where it regulates gene expression and chromatin structure. TSPYL2 acts as an inhibitor of cell-cycle progression and is involved in transcriptional repression, notably suppressing oncogenic targets by functioning within complexes such as REST/NRSF and interacting with factors like CBP/p300. Its activities include chromatin remodeling, enhancing p53-dependent apoptosis after DNA damage, and modulating signaling pathways (including TGF-β). TSPYL2 plays a crucial role in suppressing tumor growth and maintaining normal epithelial homeostasis. It is involved in neuronal gene regulation, memory formation, and is linked to neurodevelopmental disorders, cutaneous T-cell lymphoma antigens, and cognitive variability. Despite these roles, TSPYL2 is not a direct receptor, enzyme, channel, or transporter, and currently lacks interacting drugs or defined clinical biomarkers.
