Testis-specific Y-encoded-like protein 4 (TSPYL4) is a member of the nucleosome assembly protein (NAP) family, encoded by the TSPYL4 gene (HGNC: 21559, UniProt: Q9UJ04)[3][5]. It is predicted to bind chromatin and histones, participate in nucleosome assembly, and localize primarily to the nucleus and chromatin[1][3]. TSPYL4 is involved in the regulation of gene expression, notably as a negative regulator of the Wnt/β-catenin signaling pathway and as a modulator of cytochrome P450 (CYP) genes, which are important for drug metabolism and cholesterol homeostasis[2]. Diseases associated with TSPYL4 include idiopathic generalized epilepsy and generalized epilepsy with febrile seizures plus, type 6[3]. Though TSPYL4 is implicated in multiple cellular processes and disease contexts, there are no current reports of approved drugs that directly target this protein, nor is it listed as a classic therapeutic target such as a receptor, ion channel, or enzyme[3].
