Testis-specific Y-encoded protein 1 (TSPY1) is a member of the SET/NAP (TTSN) superfamily, encoded by a multicopy gene on the human Y chromosome and expressed predominantly in adult human spermatogonia[1][4][6]. It functions primarily in spermatogenesis, promoting spermatogonial proliferation via regulation of the cell cycle, suppression of USP7-mediated p53 function, maintenance of spermatogonial homeostasis, and chromatin remodeling[1][4]. TSPY1 acts as a cancer/testis antigen, is strongly upregulated in gonadoblastoma and certain cancers (testis, prostate, liver), and may contribute to tumorigenesis when aberrantly expressed in somatic tissues[1][5][6]. Copy number variations of TSPY1 are associated with male infertility and cancer susceptibility[1].
