Overview

Tetanus toxoid-specific B-cell and T-cell receptors (BCRs and TCRs) are the specialized immune receptors that recognize and respond to the tetanus toxoid (TT), an inactivated form of the neurotoxin produced by Clostridium tetani. These receptors are central to the adaptive immune response, where BCRs on B cells bind directly to the toxoid to initiate antibody production, and TCRs on CD4+ T cells recognize TT-derived peptides presented by MHC class II molecules to provide essential T-cell help. This interaction leads to the generation of high-affinity IgG antibodies and long-lived memory cells, which provide decades of protection against tetanus. Beyond their role in preventing tetanus, these receptors are frequently utilized in clinical research and vaccine development as a model for understanding immunological memory and as targets for carrier-protein-based vaccines. Tetanus toxoid is a common carrier protein in conjugate vaccines, where it recruits TT-specific T-helper cells to enhance the immune response against poorly immunogenic antigens, such as bacterial polysaccharides.
