Overview

Tetrahydrobiopterin-dependent enzymes are a group of enzymes that require tetrahydrobiopterin (BH4) as an essential cofactor for their catalytic activity. BH4 is present in nearly every cell or tissue of higher organisms and plays a central role in several critical metabolic pathways, particularly those involving amino acid metabolism and neurotransmitter synthesis. Key enzymes include phenylalanine hydroxylase (PAH), tyrosine hydroxylase (TH), tryptophan hydroxylase (TPH), and nitric oxide synthases (NOSs). They are involved in synthesis of monoamine neurotransmitters, metabolism of phenylalanine, and production of nitric oxide. Deficiencies or dysfunctions can result in severe neurological disorders and are implicated in cardiovascular diseases.
