Overview

Tetraspanin-32 (TSPAN32) is a member of the tetraspanin superfamily, a group of four-pass transmembrane proteins that play roles in cell-cell interactions, membrane organization, and signal transduction, particularly in hematopoietic tissues[1][2]. TSPAN32 is considered a tumor suppressor and is located in the imprinted gene domain of chromosome 11p15.5, a region frequently implicated in several cancers and Beckwith-Wiedemann syndrome[1][4]. Its expression is primarily confined to hematopoietic cells, and it is involved in regulating processes such as proliferation, apoptosis, immune response, and maintenance of hematopoietic function[1][2][4]. Unlike many tetraspanins, Tetraspanin-32 exhibits tumor-suppressive actions: its overexpression can inhibit leukemia progression and the proliferation of leukemia stem cells[2]. Alterations in the TSPAN32 gene or its regulatory region are associated with increased susceptibility to cancers and hematological disorders[1][4].
