Tetraspanin-33 (TSPAN33) is a member of the tetraspanin family of four-pass transmembrane proteins, primarily localized to the plasma membrane, where it functions as a scaffold protein and regulator of cell–cell interactions and signal transduction[3][4][1]. In humans, TSPAN33 expression is strongly induced in activated B cells and several B cell-related lymphomas, while it is low or absent in resting B cells. It is also expressed in certain autoimmune disease states, suggesting a role in B cell-mediated immune pathology. Beyond hematopoietic tissues, TSPAN33 plays a role in erythroid progenitor differentiation and may interact with the metalloprotease ADAM10, influencing protein maturation and trafficking. TSPAN33 is under investigation as a diagnostic and therapeutic target, particularly for malignancies and autoimmune diseases involving pathogenic B cell activation. No approved therapeutic agents currently target TSPAN33 directly[2][3].
