Tetraspanin-9 (TSPAN9) is a multi-pass transmembrane protein belonging to the tetraspanin (transmembrane 4 superfamily) family, characterized by four hydrophobic transmembrane domains and two extracellular loops with a distinctive cysteine pattern[2][3][4]. Tetraspanin-9 is widely expressed and primarily localizes to the plasma membrane, where it colocalizes with key signaling and adhesion molecules within specialized membrane microdomains. It regulates processes such as cell surface organization, signal transduction, and cell motility, and is implicated in platelet function, in part through its colocalization with GP6 in platelets[4][5]. In cancer, overexpression of Tetraspanin-9 has been shown to inhibit cell proliferation, at least in gastric cancer models[1]. To date, no drugs target Tetraspanin-9 directly, and it is not an established therapeutic or diagnostic biomarker.
