Overview

Tetratricopeptide repeat domain 16 (TTC16) is a nuclear, uncharacterized protein encoded by the *TTC16* gene on human chromosome 9q34.11[1][7]. The protein contains ten tetratricopeptide repeat (TPR) motifs—structural features typically mediating protein-protein interactions—and a Pumilio-family domain at its C-terminus[1]. TTC16 has two main isoforms and 17 transcript variants; the protein is 873 amino acids and predicted to be basic (pI 9.15)[1]. TTC16 is highly conserved across species, but has no known paralogs in humans[1][7]. Expression is highest in testis, with notable expression in lung, pituitary gland, tonsil, omental adipose tissue (especially in obese children), and immune cells such as CD8+ cells[1]. Although its precise function is not defined, the association with TPR motifs and tissue expression patterns suggest a likely role in immune system response, protein-protein interaction stabilization, and possibly transcriptional regulation[1][2]. Only one interaction partner has been identified: polymerase basic protein 2, involved in viral transcription initiation[1]. TTC16 is not currently known as a therapeutic target, disease gene, or clinical biomarker. No drugs or therapeutic strategies are associated with this gene, and its involvement in human disease remains speculative[1][7].
