Overview

Tetratricopeptide repeat domain 22 (TTC22) encodes a protein containing seven tetratricopeptide repeat (TPR) motifs that mediate protein-protein interactions and the assembly of multiprotein complexes. TPR-containing proteins act as scaffolds in a range of cellular pathways, including RNA metabolism and gene expression regulation. TTC22 has been implicated in regulating m6A-mediated gene expression and is associated with cancer progression, particularly enhancing metastasis in colon cancer through stabilization and upregulation of WTAP mRNA, as well as promoting migration and invasion in pancreatic cancer cells. High TTC22 expression correlates with poor survival in colon and pancreatic cancers and exhibits relationships with immune cell infiltration—particularly a negative association with plasmacytoid dendritic cell populations in pancreatic adenocarcinoma. TTC22 is also associated with variation in low-density lipoprotein cholesterol at the genetic and transcriptomic levels, suggesting a broader biological role beyond oncology. Alternative splicing variants exist for this gene, possibly reflecting diverse functions in different tissues. No direct drug interactions or specific mechanisms of drug action have been identified for TTC22 to date, but its biological roles and prognostic utility in multiple cancers make it a potential novel therapeutic and diagnostic target.
