Overview

Tetratricopeptide repeat domain 32 (TTC32) is a protein-coding gene that encodes a member of the tetratricopeptide repeat (TPR) domain-containing protein family, characterized by their role as scaffolds for protein-protein interactions[1][3]. While the specific molecular function of TTC32 remains incompletely characterized, it is associated with maintenance of Golgi apparatus function and protein trafficking, the disruption of which can result in neuronal dysfunction, fragmentation of the Golgi, and impaired synaptic connectivity in cerebellar circuits[1]. Genomic studies also link variants at the TTC32-WDR35 locus with increased risk for coronary artery disease[1]. Transcriptomic studies show altered TTC32 expression in various cancers, including esophageal squamous cell carcinoma and endometrial carcinoma, suggesting a possible contribution to malignant transformation[4]. However, as of now, TTC32 is not established as a direct therapeutic target, there are no drugs known to target it, and its precise cellular and disease mechanisms require further elucidation[1][4][3].
