Overview

Tetratricopeptide repeat domain 33 (TTC33) is a protein in humans encoded by the TTC33 gene, characterized by the presence of tetratricopeptide repeat (TPR) motifs—a structural scaffold of tandem 34-amino-acid repeats that mediate protein–protein interactions[1][8]. TTC33 belongs to the broad TPR protein family, which facilitates formation of multiprotein complexes, but there is presently limited direct evidence for its involvement as a receptor, enzyme, transporter, or other therapeutic drug target[3][8]. TTC33 is associated with mixed fibrolamellar hepatocellular carcinoma, although its role in disease etiology and biological function remains to be clarified[3]. No interacting drugs or biomarker roles are described, nor are safety concerns related to therapeutic interventions targeting TTC33. Current understanding suggests TTC33's primary function is scaffolding within multiprotein complexes rather than direct signaling, metabolic, or gene regulatory activity[1][8].
