TFAP2A antisense RNA 1 (TFAP2A-AS1, also known as MIR5689HG) is a long non-coding RNA transcribed antisense to the TFAP2A gene. It does not encode a protein but regulates gene expression post-transcriptionally, often by serving as a molecular sponge (competing endogenous RNA) for specific microRNAs. Overexpression of TFAP2A-AS1 is observed in several cancers, including non-small cell lung cancer and oral squamous cell carcinoma, where its increased levels are associated with enhanced tumor cell proliferation, invasion, and migration. Mechanistically, in NSCLC, it contributes to oncogenesis in part by modulating the miR-548a-3p/CDK4 axis, while in OSCC it may intersect with the p53 signaling pathway. Its expression in tumors can have diagnostic value, but it is not currently a therapeutic target, nor are there approved drugs or therapies directed against it[2][4][5].
