TGFB-induced factor homeobox 1 (TGIF1) is a highly conserved transcriptional regulator belonging to the homeobox protein family (specifically, the TALE superclass), which functions primarily as a repressor of gene transcription activated by TGF-β signaling. TGIF1 inhibits transcriptional activation by interacting with retinoid X receptor response elements and serving as a corepressor for SMAD2, linking TGF-β/nodal signaling to forebrain and midline development. Mutations or dysregulation of TGIF1 are associated with holoprosencephaly (a major developmental brain disorder), various cancers (including glioma and acute myeloid leukemia), and other conditions such as ocular disease and psychosis. TGIF1 is essential for proper embryonic development and for modulating cell differentiation, proliferation, and migration in multiple tissues. While it is not a current primary therapeutic drug target, its central role in developmental biology and disease progression makes it of substantial interest in biomedical research
