Overview

TGFB2 overlapping transcript 1 (TGFB2-OT1), also known as FLJ11812, is a long non-coding RNA (lncRNA) derived from the 3′ untranslated region (UTR) of the TGFB2 gene. Unlike protein-coding targets, TGFB2-OT1 does not encode a protein but exerts function by acting as a competing endogenous RNA (ceRNA), binding microRNAs such as miR-4459, miR-3960, and miR-4488 to regulate the availability and expression of their downstream targets (including ATG13, CDC20B, CERS1, LARP1). TGFB2-OT1 is implicated in the regulation of autophagy, cell cycle, and maintenance of embryonic stem cell stemness, and is upregulated in response to cellular stress and inflammation. Its dysregulation can influence processes such as cancer progression and embryonic development, but it is primarily studied as a molecular regulator, not as a direct therapeutic drug target.

Mechanism of action

Inhibition by small molecules (e.g., 3BDO downregulates TGFB2-OT1 RNA); RNA–miRNA interaction: acts as a molecular sponge for specific miRNAs (e.g., miR-4459, miR-3960, miR-4488), indirectly modulating miRNA target genes (e.g., ATG13, CDC20B, CERS1, LARP1)
