Overview

TGFB3 antisense RNA 1 (TGFB3-AS1) is a long non-coding RNA transcribed antisense to the TGFB3 (transforming growth factor beta 3) gene. Unlike protein-coding therapeutic targets, it functions primarily through post-transcriptional regulation: TGFB3-AS1 regulates inflammatory response in macrophages by inhibiting the maturation of miR-144, a microRNA, and stabilizing Rap1a, a signaling protein. These actions promote Wnt signaling and inflammatory cytokine expression, particularly in contexts such as homocysteine-induced macrophage activation. Currently, there are no known drugs that directly target TGFB3-AS1, nor is it a recognized therapeutic target, but it is implicated in disease-related inflammatory processes[2].

Mechanism of action

TGFB3-AS1 inhibits maturation of miR-144 by directly binding to its primary transcript (pri-miR-144) and interfering with Drosha and DGCR8-mediated processing[2] Stabilizes Rap1a protein via inhibition of ubiquitin-proteasome-mediated degradation, thereby augmenting Wnt signaling and promoting inflammatory gene expression in macrophages[2]
