Overview

THAP domain-containing protein 5 (THAP5) is a member of the THAP protein family, characterized by a conserved N-terminal THAP domain, approximately 90 amino acids in length, that mediates zinc-dependent sequence-specific DNA binding[1][2][3]. THAP5 appears to act as a transcriptional repressor in vitro and participates in the negative regulation of both cell cycle progression and transcription by RNA polymerase II[1][3]. It is mainly located within chromatin and nucleoplasm and likely contributes to chromatin modification and gene expression regulation[1][2][3]. While its precise biological role remains incompletely defined, THAP5, along with related THAP proteins, has been implicated in diverse cellular functions such as cell proliferation, apoptosis, chromatin modification, and transcriptional regulation[1][2][3]. Current data associate THAP5 with certain disease states, notably Townes-Brocks syndrome, dystonia, and possibly cancer, heart disease, and neurological disorders, but it is not a validated therapeutic drug target at present[2][3].
