Overview

THAP domain-containing protein 8 (THAP8) is a human protein characterized by the presence of a THAP domain—a conserved, zinc-dependent, sequence-specific DNA-binding domain belonging to the zinc-finger superfamily[1][2]. The THAP protein family is mainly restricted to animals and includes approximately a dozen members in humans, each with potentially distinct physiological functions. While the precise biological functions of THAP8 itself are not fully defined, THAP domain proteins as a group have been implicated in DNA binding, possible transcriptional regulation, and roles in cell proliferation, apoptosis, cell cycle modulations, and chromatin modification[1][2][3]. For THAP8, there is predictive evidence for roles in DNA binding and metal ion (zinc) binding[4]. Disease association has been described for primary autosomal recessive microcephaly; however, detailed molecular mechanisms and broader disease implications remain largely unknown[4]. Presently, THAP8 is not recognized as a validated therapeutic target, nor are there drugs or biomarkers specifically linked to its modulation or activity in clinical contexts[4].
