Overview

Thermogenesis pathway components refer to a collective group of proteins, receptors, and enzymes that facilitate the conversion of chemical energy into heat, primarily within brown and beige adipose tissues (NIH, 2021). The central effector is Uncoupling Protein 1 (UCP1), which uncouples mitochondrial respiration from ATP synthesis to generate heat (MDPI, 2021). This process is classically regulated by the sympathetic nervous system via beta-3 adrenergic receptors (ADRB3), but it also involves thyroid hormone receptors, PPAR gamma, and various secreted factors like FGF21 and Irisin (Frontiers, 2021). Pharmacological activation of these components, such as through ADRB3 agonists like mirabegron or GLP-1 receptor agonists, is a major area of research for treating obesity and type 2 diabetes by increasing whole-body energy expenditure (MDPI, 2021). Beyond UCP1, alternative thermogenic mechanisms involving futile creatine and calcium cycling are also being explored as therapeutic targets (Pharmaceutical Technology, 2025). However, challenges remain in achieving tissue-specific activation to avoid adverse cardiovascular effects and ensuring the maintenance of lean body mass during weight loss (NIH, 2021).
