Overview

Thiamine pyrophosphate-dependent enzymes are a diverse family of enzymes that require thiamine pyrophosphate (TPP, also known as thiamine diphosphate or cocarboxylase) as an essential cofactor for their catalytic activity. TPP is the bioactive form of vitamin B1 and is present in all living systems, where it catalyzes several key biochemical reactions involved in energy metabolism and biosynthetic pathways. These enzymes play critical roles in carbohydrate metabolism, energy production, synthesis of neurotransmitters, antioxidants, and pentose sugars. Notable examples include pyruvate dehydrogenase complex, alpha-ketoglutarate dehydrogenase complex, branched-chain alpha-keto acid dehydrogenase complex, transketolase, and pyruvate decarboxylase.
