Thiamine pyrophosphate (TPP), also known as thiamine diphosphate (TDP), is the biologically active coenzyme derivative of vitamin B1 (thiamine), essential for cellular energy metabolism across all domains of life [1][2]. It is synthesized intracellularly from thiamine by the enzyme thiamine pyrophosphokinase and serves as a critical prosthetic group for several multi-enzyme complexes, including pyruvate dehydrogenase and alpha-ketoglutarate dehydrogenase [1][4]. These enzymes facilitate the oxidative decarboxylation of alpha-keto acids, bridging glycolysis with the citric acid cycle to generate ATP [2][5]. TPP is also the sole cofactor for transketolase in the pentose phosphate pathway, which is vital for producing NADPH and the pentose sugars required for nucleic acid synthesis [4][6]. In clinical medicine, TPP availability is central to treating deficiency syndromes such as Beriberi and Wernicke-Korsakoff syndrome, which manifest as severe neurological and cardiovascular dysfunction [4][6]. Beyond its enzymatic role, TPP functions as a key regulatory ligand for TPP riboswitches found in bacterial mRNA [7][8]. These riboswitches control the expression of genes involved in the biosynthesis and transport of thiamine, making them high-priority targets for the development of novel antibacterial agents [8][9]. Small-molecule analogs of TPP are currently being explored as inhibitors that bind these riboswitches to disrupt bacterial metabolism and overcome conventional antibiotic resistance [9].
Acts as a catalytic coenzyme providing a nucleophilic thiazolium ring to stabilize carbanion intermediates in decarboxylation and ketol transfer reactions; also functions as a regulatory ligand for bacterial riboswitches to modulate gene expression.
