Overview

Thioesterase superfamily member 4 (THEM4), also known as CTMP (carboxyl-terminal modulator protein), encodes an enzyme with acyl-CoA thioesterase activity that hydrolyzes medium- and long-chain fatty acyl-CoA thioesters (C14–C18), suggesting a role in mitochondrial fatty acid metabolism. THEM4 is best known for its regulatory function as a negative modulator of protein kinase B (AKT1/PKB) by inhibiting its phosphorylation at the C-terminal, thereby impacting signal transduction pathways involved in cell survival and apoptosis. The expression of THEM4 is commonly reduced in various cancers, including glioblastomas, and it has been implicated in apoptotic signaling and disease progression[1][2][4][6].
