Overview

Thioredoxin domain-containing protein 11 (TXNDC11) is a member of the protein disulfide isomerase family and contains thioredoxin-like and EF-hand-binding domains[4][3][5]. It is predicted to reside in the endoplasmic reticulum membrane and function mainly as a redox regulator, aiding in the folding of proteins (notably DUOX1 and DUOX2, components of thyroid H₂O₂ generation) but is not sufficient by itself to mediate H₂O₂ production[4][3][5]. TXNDC11 expression is induced by ER stress through the unfolded protein response element, particularly via the IRE1–sXBP1 pathway[1]. It plays a role in maintaining ER homeostasis by regulating the stability of key ER stress sensors such as ATF6α, CREB3, and CREB3L2[1]. TXNDC11 is expressed in many tissues, notably at higher levels in the thyroid and prostate[3][8]. Clinically, elevated TXNDC11 expression has been associated with poor prognosis in several cancers, including glioma (where it is linked to increased tumor grade and immune pathway activity), while correlations with prognosis may vary by cancer type[2]. No drugs or mechanisms of therapeutic modulation for TXNDC11 have been established to date, but its expression is emerging as a potential biomarker, especially for cancer prognosis[2].
