Overview

Thioredoxin domain-containing protein 3 (NME8) is a protein mainly expressed in testis and related tissues, vital for sperm tail maturation and proper ciliary function[1][3][4][5][7][8]. It contains an N-terminal thioredoxin domain and three C-terminal nucleoside diphosphate kinase homology domains, but the enzyme lacks canonical NDK activity. NME8 exhibits 3'-5' exonuclease activity preferentially on single-stranded DNA, implicating it in DNA proofreading and repair. Mutations in NME8 are causative for primary ciliary dyskinesia type 6, a disorder affecting cilia motility that presents as respiratory tract infections, infertility, and laterality defects[1][3][4][5]. NME8 is not currently recognized as a therapeutic drug target, nor are there known interacting drugs or specific safety concerns associated with modulation of its function.
