Overview

Thioredoxin (TXN) mRNA is the transcript responsible for the synthesis of thioredoxin-1, a ubiquitous redox-active protein essential for maintaining cellular thiol-disulfide balance [1]. By reducing oxidized proteins and scavenging reactive oxygen species, the TXN system protects cells from oxidative stress-induced damage and regulates various signaling pathways related to growth and survival [2]. In many malignancies, TXN mRNA is overexpressed, which correlates with tumor progression, metastasis, and resistance to apoptosis-inducing therapies [3]. Therapeutic targeting of TXN mRNA via antisense oligonucleotides or siRNA aims to silence the expression of the thioredoxin protein, thereby increasing intracellular oxidative stress and promoting cancer cell death [4]. This approach is particularly relevant in cancers that have become dependent on antioxidant pathways to survive high metabolic rates. Beyond oncology, TXN mRNA levels are studied in the context of inflammatory and cardiovascular conditions where oxidative stress plays a pathogenic role [5]. Challenges in targeting TXN mRNA include ensuring tissue-specific delivery and avoiding the systemic suppression of redox defenses necessary for normal physiological function [6].
