Overview

THO complex subunit 5 (THOC5) is an evolutionarily conserved protein that is a component of the THO subcomplex within the transcription/export (TREX) complex. It participates in coupling transcriptional elongation, mRNA 3′ end processing, and nuclear export of a selective subset of mRNAs, particularly those rapidly induced by extracellular signals such as growth factors and cytokines. THOC5 is critical for the export of immediate-early genes and is essential for stem cell survival and differentiation. Its depletion affects the processing and export of inducible transcripts but not the bulk of steady-state mRNAs. THOC5 is phosphorylated in response to various stimuli, including oncogenic events, and may play a role in cancer development. Genetic studies indicate that it is indispensable for normal stem cell function, and its loss leads to defects in cell differentiation and survival mechanisms[1][2][3][4][6].
