Overview

Threonine synthase-like 2 (THNSL2) is a human enzyme encoded by the THNSL2 gene. It acts as a catabolic phospho-lyase, degrading O-phospho-threonine and structurally related substrates to produce alpha-ketobutyrate, phosphate, and ammonia. In mouse, an alternatively spliced form has been shown to function as a cytokine called SOFAT (secreted osteoclastogenic factor of activated T cells), promoting IL-6 production in osteoblasts and potentially modulating inflammatory responses. While multiple transcript variants exist and some functional data come from animal studies, the principal characterized activity in humans is enzyme-based amino acid catabolism. There are currently no approved therapeutic drugs targeting this protein, and its direct role in human pathophysiology or as a drug target remains undetermined.
