The thrombopoietin gene promoter is a regulatory DNA sequence located upstream of the human thrombopoietin (THPO) gene, which governs the initiation and rate of TPO transcription. The THPO promoter region lacks a TATA box, showing multiple transcription start sites over a span of about 50 nucleotides and several functional cis-acting elements, including Ets family binding motifs important for high-level gene expression in the liver[6]. Genetic variants and alternative promoters (P1a and P1) within this region can influence TPO production and are associated with differences in TPO activity and disease susceptibility, particularly in cardiovascular and hematological disorders[3][5][6]. However, the promoter itself is not a conventional therapeutic target, nor does it function as a receptor, transporter, or enzyme.
