The thrombopoietin receptor, also known as MPL or CD110, is a cell surface protein primarily responsible for mediating the effects of thrombopoietin (TPO), the chief cytokine regulating platelet production and megakaryocyte development. It plays a critical role in hematopoiesis, particularly in maintaining hematopoietic stem cells and controlling platelet counts.
Thrombopoietin receptor agonists (TPO-RAs) mimic TPO action by activating the MPL receptor, stimulating increased platelet production through downstream signaling cascades involving JAK/STAT, MAPK, and PI3K-AKT pathways.
