Overview

Thrombospondin 1 (THBS1) mRNA encodes a large, multifunctional matricellular glycoprotein that serves as a critical regulator of the tumor microenvironment and tissue remodeling. THBS1 is a potent endogenous inhibitor of angiogenesis and a primary activator of latent transforming growth factor-beta (TGF-beta), positioning it as a central player in both tumor suppression and the progression of fibrotic diseases. In many malignancies, THBS1 expression is suppressed by oncogenic signaling to facilitate neovascularization, whereas its overexpression is a hallmark of fibrotic conditions such as systemic sclerosis and pulmonary hypertension. Targeting THBS1 at the mRNA level using RNA interference (siRNA) or antisense technologies is an emerging therapeutic strategy aimed at downregulating its pro-fibrotic and pro-inflammatory signaling in pathological states. Furthermore, THBS1 mRNA levels are utilized as sensitive pharmacodynamic biomarkers in clinical trials to monitor the efficacy of anti-fibrotic agents and TGF-beta inhibitors.
