A thrombospondin receptor modulator refers to any molecule or agent that influences the activity of receptors for thrombospondins (TSPs). TSPs are a family of secreted glycoproteins that play key roles in cell-cell and cell-matrix interactions, angiogenesis, synaptogenesis, inflammation, tissue repair, and tumorigenesis. Major known thrombospondin receptors include CD36, CD47 (Integrin-associated protein), αvβ3 Integrin, Syndecans, α2δ-1 Subunit of Voltage-Gated Calcium Channels, and Neuroligin 1. Modulation of these receptors can affect angiogenesis, platelet aggregation, tumorigenesis, synaptogenesis, regulation of blood flow via nitric oxide signaling pathways, and activation/inhibition of growth factors such as TGF-beta. Because these pathways are implicated in cancer progression, neuroregeneration, cardiovascular disease, fibrosis, wound healing, etc., modulators targeting thrombospondin-receptor interactions are being explored for therapeutic use.
