Thrombospondin type-1 domain-containing protein 1 (THSD1) is an extracellular matrix-associated transmembrane protein containing a thrombospondin type 1 domain, primarily involved in the regulation of vascular homeostasis by influencing focal adhesion dynamics and endothelial cell function[1][3]. It acts as a positive regulator of nascent focal adhesion assembly, modulates attachments between endothelial cells and the extracellular matrix, and maintains adhesion stability through interactions with focal adhesion kinase (FAK)[3]. THSD1 also negatively regulates autophagy processing at focal adhesions by modulating Beclin 1 phosphorylation, thus preventing excessive focal adhesion turnover and promoting proper barrier integrity in brain endothelial cells[1]. Mutations or epigenetic silencing of THSD1 contribute to vascular diseases (such as intracranial aneurysm and subarachnoid hemorrhage) and may contribute to tumorigenesis through loss of its tumor suppressor activity[1][3]. Multiple alternatively spliced transcript variants exist encoding different isoforms[2][3]. No drugs or drug mechanisms specifically targeting THSD1 are currently known, and it is not yet defined as a classic therapeutic target[3].
