Overview

The thromboxane A₂ receptor (TP) is a seven-transmembrane G protein-coupled receptor encoded by the TBXA2R gene, primarily mediating the cellular and physiological effects of thromboxane A₂ (TXA₂), a bioactive lipid derived from arachidonic acid. Activation of the TP receptor plays a critical role in platelet aggregation, vasoconstriction, and overall hemostasis, making it fundamental in both normal vascular function and the pathogenesis of diseases such as thrombosis and atherosclerosis[1][2][3][7][10]. The receptor exists as two human isoforms, TPα and TPβ, which differ in cellular distribution and downstream signaling mechanisms[1][2]. TP signaling is mediated by heterotrimeric G proteins, leading to diverse cellular responses. TP antagonists and drugs inhibiting TXA₂ synthesis are primary pharmacological strategies for antiplatelet therapy and cardiovascular disease prevention[3][10]. Overactivation or underactivation of the TP receptor can result in pathological bleeding or excessive clotting[1], and the TP pathway is also implicated in inflammation and cancer, particularly breast cancer progression[8].
